Is there a comparison document that anyone is aware of between Application Server and Access Essentials?

Or if anyone can give the key differentiators between them that would be ideal

Well they are different products. Citrix Access Essentials is based on ICA; 2X ApplicationServer uses the built-in Microsoft RDP protocol.
Citrix AE includes a TS CAL on its licensing costs; 2X AS does not, this is extra (but even with this extra cost the total solution cost will be MUCH cheaper).
Citrix AE is limited to 75 users on a single server. 2X AS has no limitations. You can even use it with the 2X LoadBalancer to create a load balanced farm with 2, 3, 10 servers for hundred of users.
The main thing is to understand what your requirements are and why you thought about Citrix. Then we will be able to tell you what the best way to go is.

Currently, you can't use the 2x load balancer to load balance a farm of 2x application servers. Apparently that will be coming soon though.

Oh yes, you can use the LoadBalancer to load balance ApplicationServer. There are some caveats but it is possible and yes, it works.
