Getting 2x Application Server to work through ISA server

Hi.

We hav set up a test enviroment with 2x Applicationserver. We recently upgraded the system to the 4.1 release, and then everything stoped working. All clients connection on the same subnet as the 2x server have no problems, but all users on subnets that are filtered by the ISA server and all users on VPN connections can't connect to the server.

What have changed? What do I need to open in the ISA server to get it to work?
